I'm asking about the Amsterdam to Vienna cruise, Rhine, Mainz, Danube
rivers, in May.

Generally yes. For instance in Amsterdam it is close to downtown (walking
distance but hardly central. In smaller cities the river ships tie up in
the heart of town.

Sounds like the Uniglobe cruise we took last September (we went the
other way); if so then yes, usually. If you don't want to take the
planned excursion, you can either walk right into town or pick up a bus
or taxi pretty much at dockside.

Once, they docked briefly before reaching town (in the middle of nowhere
at some sort of industrial pier facility), took us into town by bus,
picked us up later and then met the ship which has actually sailed past
town and further downstream. In that instance, if you don't want to take
the tour, you're pretty much stuck.
